Frequently asked questions
Lentiviral or retroviral vector — how do I choose?
Gamma-retroviral vectors such as Retro-X integrate only in actively dividing cells, because the pre-integration complex cannot cross an intact nuclear envelope. Lentiviral vectors like Lenti-X integrate in dividing and non-dividing cells, which is why primary neurons, macrophages and quiescent stem cells are handled lentivirally. Where the model divides rapidly and a simpler system is enough, a retroviral vector remains a sound choice.
How do I measure lentiviral titer?
Two routes serve different moments. Lenti-X GoStix Plus is a 10-minute lateral-flow test on 20 µl of supernatant that reads p24 and is quantified through a free smartphone app — useful for checking a packaging run before committing target cells. The Lenti-X qRT-PCR Titration Kit quantifies viral RNA genomes in about four hours and works with all HIV-1-based lentiviral vectors.
What does a fourth-generation packaging system mean in practice?
In the Lenti-X system the packaging and non-viral components are split across a suite of five vectors, premixed at an optimized ratio and transfected into Lenti-X 293T cells. Spreading the genome across more plasmids lowers the chance that recombination regenerates a replication-competent virus, and the premix removes ratio optimization from your bench work. Takara reports an average titer of 1.85 x 10^8 IFU/ml for pLVX vectors packaged this way.
Which fluorescent protein — a marker or a fusion tag?
For fusion constructs choose a true monomer, so the tag does not impose artificial oligomerization: AcGFP1, mCherry and DsRed-Monomer fall in that group. For tracking transduction or transfection efficiency, brightness outweighs oligomeric state, and ZsGreen1 or tdTomato are the usual picks — tdTomato is roughly six times brighter than EGFP and emits at 581 nm, which suits live-animal imaging. Bicistronic pLVX-IRES vectors express your gene and the marker from a single transcript.
Can I run a reporter assay without lysing the cells?
Yes, using secreted reporters. The Ready-To-Glow assay measures secreted Metridia luciferase directly in the culture medium, so one well can be sampled repeatedly across a time course; signal is generally detectable 12–18 hours after transfection and peaks between 48 and 72 hours. The dual version adds secreted alkaline phosphatase (SEAP) as a second reporter or a normalization control.
Who supplies Lenti-X and Retro-X in Israel?
Takara Bio's viral transduction and reporter portfolio is carried for Israeli labs by Danyel Biotech as official Israeli distributor, with cold-chain delivery of packaging mixes and producer cell lines. Viral vector work stays subject to your institution's biosafety rules; talk to us for a quote or for help sizing a packaging run.
