Frequently asked questions

What is the difference between sequencing STRs and typing them by capillary electrophoresis?

Capillary electrophoresis reports an STR allele by its length. Sequencing reads the repeat itself, so two alleles of the same length can be told apart when their sequence differs, and STRs and SNPs can be run in one library. The ForenSeq Signature Plus Kit covers more than 200 markers in a single workflow, most SNP amplicons are 125 bp or shorter, and its allele calls remain compatible with existing CE nomenclature.

Which ForenSeq kit is meant for routine database casework?

ForenSeq MainstAY targets 27 autosomal STRs, 25 Y-STRs and Amelogenin in one reaction and is built for high-volume casework, up to 96 samples per run on the MiSeq FGx system. The MainstAY SE version adds the SE33 marker for laboratories whose national database requires it.

What is forensic investigative genetic genealogy, and what does the Kintelligence kit do?

Forensic investigative genetic genealogy (FIGG) searches a DNA profile against genealogy databases to find relatives and build a family tree that points to a candidate. The ForenSeq Kintelligence Kit targets 10,230 SNPs selected for kinship and leaves out medically informative markers. Results are reported in a format for GEDmatch PRO. The Kintelligence HT Kit uses the same SNPs, runs up to 12 post-mortem or 36 ante-mortem libraries per run, and adds a local kinship database for identifying human remains.

When is mitochondrial DNA sequencing used instead of STR typing?

Mitochondrial DNA is present in many copies per cell and is inherited from the mother, so it can give a result from hair shafts, old bone and other samples with too little nuclear DNA for STRs. The ForenSeq mtDNA Control Region Kit covers about 1,200 bp of the control region in 18 overlapping amplicons from 100 pg of DNA. The mtDNA Whole Genome Kit tiles overlapping amplicons across the entire mitochondrial genome for higher resolution on degraded samples.

Can the system estimate appearance or ancestry from DNA?

The ForenSeq Imagen Kit estimates eye colour and skin colour from SNP sets established by forensic laboratories, and a second primer mix adds biogeographical ancestry. These results are investigative leads, not identifications, and the ancestry option is used only where local regulation permits.

Who supplies the MiSeq FGx system and ForenSeq kits in Israel?

Danyel Biotech is the official Israeli distributor of QIAGEN's Human ID and forensics range, including the kits developed by Verogen. Installation and application support are handled locally. Request a quote from any product page.

Buying Guide

Start from the question the case asks. For STR profiles that go to a national database, use ForenSeq MainstAY, or the SE version when SE33 is required. For mixtures, degraded samples, or STRs and SNPs in one run, use ForenSeq Signature Plus. For kinship and genealogy leads, use Kintelligence, and Kintelligence HT when identifying remains at scale. For hair shafts and old bone, start with the mtDNA Control Region Kit and move to the Whole Genome Kit when more resolution is needed. Appearance and ancestry leads come from Imagen. Every kit runs on the MiSeq FGx system with the MiSeq FGx Reagent Kit, or the Micro Kit for small runs.
