Frequently asked questions
Why treat RNA with DNase I before reverse transcription?
Residual genomic DNA is amplified by any primer pair that does not span an intron, which inflates the apparent transcript level. RNase-free DNase I hydrolyses both double- and single-stranded DNA into short oligonucleotides and mononucleotides, clearing that carry-over before the RT step. Inactivate or remove the enzyme afterwards so it cannot act on downstream reactions.
What is Exonuclease I used for?
Exonuclease I from E. coli is a magnesium-dependent 3'-to-5' exonuclease specific for single-stranded DNA, so it degrades leftover PCR primers while the double-stranded product is untouched. Paired with a phosphatase to remove unincorporated dNTPs, it is the standard enzymatic cleanup ahead of Sanger sequencing — no columns and no elution losses.
Which enzymes does an in vitro transcribed mRNA workflow need?
Typically a DNase to clear the DNA template, an RNase inhibitor such as RiboGuard to protect the transcript through long incubations, plus capping or ligation steps depending on the construct design; LGC groups this set in its IVT mRNA enzyme resource. A separate case is circularising single-stranded DNA, which is what CircLigase ssDNA Ligase is for.
Who supplies Lucigen and Biosearch enzymes in Israel?
These enzymes reach Israeli labs through Danyel Biotech, official Israeli distributor for LGC Biosearch Technologies. Lucigen and Epicentre products are now sold under the Biosearch Technologies name, so older catalog numbers still map onto the current range — send yours across and we will quote against them.
