Frequently asked questions
What is the difference between PCR, RT-PCR, qPCR, and RT-qPCR?
PCR amplifies DNA; qPCR (also called real-time or quantitative PCR) measures amplification as it happens. RT-PCR first converts RNA to cDNA using reverse transcription, and RT-qPCR does that quantitatively in real time. RT means reverse transcription — not real-time — which is the source of most confusion.
Is TB Green the same as SYBR Green?
Functionally yes for most users — TB Green is Takara's name for its intercalating green dye. In December 2017 Takara renamed its SYBR Premix Ex Taq line to TB Green Premix; catalog numbers and performance stayed the same.
Should I choose a dye-based or probe-based qPCR mix?
Dye-based chemistry (TB Green/SYBR) is economical and flexible — ideal for screening and gene-expression work with well-designed primers. Probe-based chemistry (TaqMan-style, 5' nuclease) adds sequence-specific detection, needed for multiplexing and diagnostic-grade specificity.
When do I need a hot-start polymerase?
When reactions are set up at room temperature or amplify low-copy targets. Hot-start enzymes such as Takara Taq HS are blocked (typically by an anti-Taq antibody) until the first denaturation step, preventing mis-priming and primer dimers without changing fidelity.
Should I run one-step or two-step RT-qPCR?
One-step kits combine reverse transcription and qPCR in a single tube — faster, fewer pipetting steps, less contamination risk. Two-step workflows create a cDNA bank you can revisit to test multiple genes from one sample. For two-step workflows, see our Reverse Transcription & cDNA Synthesis category.
