HEK 293 Tet-Off Advanced is a transformed human embryonic kidney-derived cell line that expresses the tetracycline (Tet)-regulated transactivator Tet-Off Advanced. Inducible expression of any gene can be achieved by transfecting or transducing this cell line with a vector containing your gene of interest under the control of a tetracycline-responsive promoter. Expression is induced by the withdrawal of doxycycline (Dox) from the culture medium. Tet-inducible cell lines are shipped with Tet System Approved FBS and a control cell line created for inducible expression of luciferase.
Features
Stably expressed Tet-Off transactivator protein in commonly used cell lines
Induce expression of your GOI by removing Dox from the culture of transfected cells
Easily create an inducible system by first placing your gene under control of a tetracycline-responsive promoter (e.g.,PTRE3G), transfecting it into one of these G418-resistant cell lines, and selecting for double-stable clones
