Frequently asked questions
What is the difference between HiTrap and HisTrap columns?
HiTrap is Cytiva's prepacked 1 mL and 5 mL column format, offered with many different resins — IMAC, heparin, NHS-activated, and more. HisTrap columns are HiTrap columns packed specifically with Ni Sepharose for His-tagged protein purification: HisTrap HP uses 34 µm beads for high resolution and binds at least 40 mg of His-tag protein per mL of resin, while HisTrap FF Crude is designed for direct loading of crude samples.
Can I run HiTrap columns without an ÄKTA system?
Yes. HiTrap-format columns are designed to run with a manual syringe, a peristaltic pump, or a chromatography system such as an ÄKTA. Syringe operation is a practical way to do method scouting, group separations, and small-scale purifications before committing to a system run.
What is the difference between Sepharose Fast Flow and High Performance resins?
Fast Flow (FF) resins use rigid, larger beads that tolerate flow rates up to 300 cm/h — the choice for capture and intermediate steps and for processing large volumes. High Performance (HP) resins use smaller beads (around 34 µm) that trade speed for sharper peaks, making them better for high-resolution and polishing work.
Which size exclusion resin should I choose — Superdex, Sephacryl, or Sephadex?
Superdex, a composite of dextran and agarose, is Cytiva's first choice for high-resolution fractionation with short run times — Superdex 75 suits most recombinant proteins (3000–70 000 Da). Sephacryl handles fast, high-recovery separations from lab to industrial scale across wide molecular weight ranges. Sephadex, the classic cross-linked dextran, is typically used for group separations such as desalting and buffer exchange.
Why choose a polymer HPLC column over silica?
Polystyrene-divinylbenzene (PS-DVB) packings like Hamilton's PRP-1 are chemically stable across an extreme pH range where silica-based phases hydrolyze and dissolve, so they survive aggressive mobile phases and NaOH cleaning. The polymer itself is the stationary phase — no coating to strip or bleed — and its different selectivity can resolve analytes that co-elute on a silica C18.
Where can I buy Cytiva chromatography columns and resins in Israel?
As the official Israeli distributor for both Cytiva and Hamilton, Danyel Biotech keeps prepacked columns, bulk resins, and polymer HPLC columns in local stock, with application support for purification and analytical work alike. Ask for a quote directly from this page.
