It is based on the translocation of the membrane phosphatidylserine (PS) from the inner face of the plasma membrane in normal status to the cell surface after apoptosis initiation. PS on the cell surface can be detected by staining with a fluorescent conjugate of Annexin V, a protein that has a high affinity for PS. The detection can be analyzed by flow cytometry or by fluorescence microscopy.
